Extraction of flavonoids using chloroforme and butanol from Silybum marianum

New antimicrobial vegetal compounds are researched from an endemic medicinal plant, widely répandue in Mediterranean region, particularly in Algeria, Silybum marianum. For that, flavonoids have been extracted using several adapted solvent systems then separated by a thin layer chromatography (TLC) on silica gel. The antimicrobiol effect of the compounds extracted was tested using disk method against the following bacteria and fungi : Staphylococcus aureus, Staphylococcus albus, Pseudomonas sp, Escherichia coli, Serratia sp ; Aspergillus sp, Penicillum sp, Candida albicans and Saccharomyces cereviseae. The highest inhibitory effect is corresponding to chloroform and butanol extracts; both extracts presented strong activity against: Staphylococcus aureus, Staphylococcus albus, Candida albicans and Saccharomyces cereviseae. The minimum bacteriostatic and bacteriocidal inhibitory concentrations against sensitive germs were determined by solid and liquid dilution methods. Determination of CMB/CMI ratio revealed a bacteriostatic action of these flavonoids. The study of thermostability revealed their resistance at cold and heat temperatures (-5°C ; 4°C ; 40°C ; 60°C and 100°C). These results highlight antimicrobial activities of flavonoids extracted from Silybum marianum, which broadens the therapeutic properties of this plant.

Abstract The goal of this work is the study and the valorisation of a medicinal plant, widely responded in Mediterranean region, particulary in Algeria, Silybum marianum. The chloroforme and btanolic solvents extracts of Silybum marianum were screened for antibiocidal and photochemical properties. Flavonoids were detected in both extracts. These extracts were active against Staphylococcus aureus and Staphylococcus albus and Candida albicans, Saccharomyces cerevisiea with a diameter depassing (15 mm). Flavonoides were separated and identificated by a thin layer chromatography (TLC) on silica gel. The TLC results permis to identify 3 different spots S1; S2; S3.The thermo stability essays revealed their resistance at cold (-5, 4 °C) and heat temperatures (40, 60 °C) during 30 min and inactivated at 100°C. These results prove antibiocidal effects of flavonoids extracted from silybum marianum, wish enlarge the therapeutic properties of this plant.
